Abstract

Litteraturstudien har som mål att reda ut vad Atopisk Dermatit hos hund (CAD) är för något, hur det diagnosticeras och hur sjukdomen kan förebyggas. Eftersom 10-15% av hundpopulationen är drabbade av sjukdomen som ofta innebär klåda och lidande för hunden är det av stort intresse att arbeta för att förebygga förekomsten av sjukdomen samt för effektiva behandlingsmetoder. Immunförsvaret, diagnosticering, miljöexponering, genetiska
faktorer och hudbarriären är områden som behandlas i studien och är faktorer som direkt eller indirekt påverkar varandra samt uppkomsten av CAD. Det genetiska arbetet är av stor vikt för förståelsen kring CAD vilket är viktigt för att hitta bra behandlingsmetoder. Genom Genomewide
association studies (GWAS) har en kandidat region påträffats innehållande potentiella regulatoriska mutationer på genen PKP2 som kodar för ett proteinet Plakophilin 2 som finns i desmosomer vilka associeras med utvecklingen av CAD hos svenska Schäferhundar. Plakophilin 2 som troligtvis påverkar hudbarriärens funktion indikerar att hundar med CAD har en försämrad hudbarriär vilket gör det möjligt för allergener att ta sig in i kroppen genom
huden och in till den underliggande vävnaden. En försämrad hudbarriär i kombination med ett immunförsvar som är hyperreaktivt kan ge typiska symtom för CAD vilka ofta innebär lidande för hunden. För att förebygga sjukdomen bör valparnas immunförsvar ges möjlighet att skapa tolerans mot allergener. Det kan ske genom miljöexponering samt att hudbarriären behandlas så att den skall kunna upprätthålla sin funktion och skydda mot intrång av allergener.

This literature study aims to sort out the term Canine Atopic Dermatitis (CAD), how it is diagnosed and how the disease can be prevented. Since 10-15% of the dog population is affected by CAD, which often means itching and suffering for the dog, it is of great interest to
work for preventing the disease and finding efficient treatment methods. The immune system, the diagnosis, environmental exposure, genetic factors and the skin barrier are areas covered in this study and they are directly and indirectly affected by each other and the appearance of CAD. The study of the genetics behind the disease is essential for understanding and very important for development of efficient treatment methods. Using Genome-wide association studies (GWAS) a candidate region containing potential regulatory mutations in the gene PKP2 encoding the desmosomal protein Plakophilin 2 was found to be associated to CAD development in German shepherd dogs. Plakophilin 2 most likely affects the skin barrier and
indicates that dogs with the disease have a subfunctional skin barrier, which enables that allergens can break through the skin and more efficiently enter the underlying tissue. A damaged skin barrier in combination with a hypersensitive immune system evokes the typical symptoms for CAD, which often means suffering for the dog. To prevent the disease the puppies immune system should be given the opportunity to create tolerance against the allergens. This can be made by environmental exposure and the skin barrier should be treated so it can maintain its function and protect against intrusion of allergens.
