Abstract

Artificiell insemination (AI) har revolutionerat reproduktionen och husdjursaveln sedan den första inseminationen gjordes 1784. När man 1949 upptäckte att spermier kunde överleva djupfrysning och upptining underlättades AI-arbetet ytterligare. Ett lyckat resultat vid användningen av AI bygger på att alla delar i den kedja av händelser fungerar från samling av sperma till insemination. En kritisk länk i kedjan är själva handhavandet av sperman vid upptining och laddning av payetten i pistoletten i samband med inseminationen. Hur upptiningsprotokollet ser ut varierar dock mellan olika länder och instruktionerna saknar ofta vetenskapliga referenser. Totalt semineras idag cirka 85% av mjölkkorna i Sverige och merparten, cirka 60%, utförs av djurägaren själv eller dennes anställda. Ibland är förutsättningarna för insemination i djurstallarna långt ifrån optimala. Detta leder till att de givna instruktionerna inte alltid följs fullt ut. I vissa fall skulle detta kunna resultera i försämrade dräktighetsresultat och nedsatt produktion.
Syftet med denna studie var 1) att jämföra några olika upptiningsprotokoll, som idag rekommenderas i olika länder inom Europa, och studera hur spermiernas livsduglighet påverkas och 2) att undersöka temperaturförändringen inuti payetterna vid upptining enligt de olika protokollen, samt 3) vid olika handhavanden i samband med laddning av pistoletten.
En payett från tre olika frysoperationer per tjur (totalt 20 tjurar) tinades för vardera av de fem olika protokoll, som vi tagit fram, baserat på de protokoll som används inom Europa. Därefter undersöktes spermiens motilitet med hjälp av en datoriserad motilitetsanalysator (sk CASA) och spermiernas membranintegritet undersöktes med hjälp av flödescytometri. Resultatet visade att det föreföll fördelaktigt för motiliteten att spermierna tinades och förvarades i +35-37˚C under en längre upptiningstid (>30s), men eventuellt negativt för membranintegriteten. Om preliminära resultat från en pågående studie rörande korrelationen mellan olika motilitetsvariabler och dräktighetsresultat appliceras på resultaten i vår studie skulle upptining enligt protokoll D (+37˚C, 45s) vara det mest fördelaktiga.
Temperaturen inuti payetten steg betydligt snabbare och sluttemperaturen var betydligt högre vid upptining för de upptiningsprotokoll som studerades än vad som tidigare angivits i svenska dokument. Temperaturen fortsatte även att stiga ytterligare några grader efter det att payetten avlägsnats från vattenbadet, vilket är av betydelse vid användning av upptiningstemperaturer över +37˚C. Temperaturen fluktuerade kraftigt efter upptiningen oavsett upptiningsprotokoll, men temperaturfluktuationerna kunde begränsas om pistolettens främre del förvärmdes före och även skyddades efter laddningen oavsett omgivningstemperatur. Det är i dagsläget oklart vilken betydelsen temperaturfluktuationerna egentligen har för spermiernas slutliga befruktningsförmåga.

Artificial insemination (AI) has revolutionized the reproduction and livestock breeding since the first insemination was done in 1784. When the discovery that sperm could survive freezing and thawing was made in 1949, it facilitated the development of applied AI work considerably. A successful result in the use of AI is depending on each single link in a long chain of events, from the collection of semen to the insemination. A critical link in the chain is the actual handling of the semen during thawing and the loading of the straw in to the insemination gun before insemination. The thawing protocols differ between countries and the instructions are not always based on scientific studies. Today about 85% of dairy cows in Sweden are inseminated and about 60% of the inseminations are performed by the owners or herd employees. The conditions and environment in the barn for this type of work is not always optimal. Inseminators may thus not follow the instructions of thawing, but make own changes which in some cases will lead to a decreased pregnancy rates and impaired fertility and production .
The objectives of this study were 1) to compare different thawing protocols, that are currently recommended in countries in Europe, and study how sperm viability is affected and 2) to investigate the temperature change inside the straw during thawing according to the different protocols, as well as 3) at various types of handling in connection with the loading of the insemination gun.
One payette from each of three freezeing operations per bull (a total of 20 bulls) were thawed for each of five different protocols based on protocols used within Europe. Various parameters of sperm motility was investigated using a computer-assisted sperm analyzer (CASA), and sperm membrane integrity was examined by flow cytometry. The results showed that it seemed advantageous for the motility of the sperm if it was thawed and stored at +35-37˚C for a longer period of time (> 30s), but possibly negative for the membrane integrity. However, applying results from an ongoing study on the correlation between different motility variables and fertility results to our study indicated that the sperm thawed according to protocol D (+37˚C, 45s) would have the best correlation.
The temperature inside the payette increased much faster and the final temperature was significantly higher during thawing according to the thawing protocols studied than claimed in previous Swedish documents. The temperature continued to rise a few degrees more after the straw was removed from the water bath, which could be of importance when using thawing temperatures above +37˚C. The temperature fluctuated markedly after thawing regardless of the thawing protocol but we could also observe that temperature fluctuations could be limited if the front part of the insemination gun was preheated before and was protected after loading regardless of the ambient temperature. At present, it is unclear what effects temperature fluctuations really have on the sperm fertilization ability.
