Abstract

I denna studie har förekomst av progesteronreceptorer i olika typer av primära juvertumörer undersökts med hjälp av immunohistokemi. Dessutom har tumörceller från carcinomcellinjen CMT-U27 odlats i olika cellodlingsmedium och exponerats för olika testsubstanser som binder till progesteronreceptorn (progesteron och RU486/mifepristone). Cellernas receptoruttryck och progesteroninnehåll utvärderades med hjälp av ELISA och tumörcellernas viabilitet analyserades med Colorimetric Cell Viability Test.
Av resultaten framgår att samtliga undersökta primära juvertumörer uttryckte progesteronreceptorer. De odlade cellerna hade högre cellviabilitet när de odlades utan fetalt bovint serum (FBS). Efter exponering med progesteron minskade tumörcellernas uttryck av progesteronreceptorer, vilket hämmades av mifepristone. Cellerna innehöll högre halter progesteron än det medium cellerna odlades i vilket kan tolkas som att de kan producera eget progesteron. De celler som inkuberades i endast progesteron var den enda grupp vars cellviabilitet minskade efter 48 timmars inkubation, vilket kan bero på att progesteron får celler att stanna i G1-fasen av cellcykeln och därmed hämma deras tillväxt.

In this study the occurrence of progesterone receptors in different mammary tumours have been examined with immunohistochemistry. Furthermore tumour cells from the carcinoma cell line CMT-U27 have been cultured in different cell culture media and exposed to different test substances that bind to the progesterone receptor (progesterone and RU486/mifepristone). The receptor expression and progesterone content of the cells have been evaluated with ELISA and the cell viability was analyzed with Colorimetric Cell Viability Test.
The results show that all of the examined primary tumours expressed progesterone receptors. The cultured tumour cells hade higher cell viability when grown without fetal bovine serum (FBS). Exposure of progesterone lowered the expression of progesterone receptors in the cells, an effect that was inhibited by mifepristone. The cells contained a higher rate of progesterone than the conditioned medium. This indicates that the cells produce their own progesterone. The cells that were incubated in progesterone alone was the only group with lower cell viability after 48 hours of incubation, this might be because of the effect of progesterone on cell cycle progression with an arrest in the G1-phase and thereby inhibiting the cell proliferation.
