Abstract

Felint leukemivirus (FeLV) är ett retrovirus som ger upphov till en rad olika sjukdomstillstånd, däribland tumörsjukdomar. Viruset kan delas in i fyra subtyper (FeLV-A, -B, -C och -T) med naturligt förekommande variationer inom subgrupperna. Idag kan vi vaccinera mot FeLV men prevalensstudier har visat att viruset inte är så utrotat som man tidigare har trott att det är.

Framförallt subtyperna FeLV-A och FeLV-B har förknippats med tumörsjukdomar. Den ledande teorin är att tumörerna uppstår på grund av den retrovirala mekanismen insertionsmutagenes vilken aktiverar signalvägar och genererar produkter förknippade med tumöruppkomst.

De styrande elementen gällande tumörform verkar vara sekvenser i virusets LTR och env gen. Specifika sekvenser i dessa områden skiljer sig åt mellan virusisolat och har associerats både naturligt och experimentellt med särskilda tumörer. I pro-viralt LTR från multicentriska lymfom har en specifik 21 basparssekvens replikerad i tandem hittats som inte bara modulerar det LTR-drivna genuttrycket utan även ger cellen en replikativ fördel. T-cellstumörer, särskilt thymuslymfom, har förknippats med pro-virala insertioner innehållandes dubblerade enhancers. Även inokulationsplats och värden själv verkar påverka sjukdomsutgången.

Forskning pågår inom ämnet och förhoppningsvis kan ny information gynna inte bara katter med tumörer orsakade av FeLV utan även andra arter drabbade av andra tumörer. Forskarna hoppas även att en ökad kunskap om FeLV ska medföra en större kännedom om andra retrovirus och deras mekanismer, som till exempel humant immunbristvirus (HIV).

The feline leukaemia virus (FeLV) is a retrovirus and the causative agent in several diseases, some of them cancerous. The virus is divided into four subgroups (FeLV-A, -B, -C and –D) within which there are natural variations. Vaccines against FeLV are available today but recent prevalence studies suggest that the virus is not as rare as it was thought to be.

FeLV-A and FeLV-B has predominantly been associated with the tumours induced by the virus. Most scientists believe that the tumours are caused by the retroviral mechanism insertional mutagenesis which activates signal pathways and generate products linked to tumourigenesis.

Sequences in the viral LTR and env gene appear to be the deciding elements concerning what kind of tumour will be formed. Specific sequences found in these areas differ between viral isolates and the sequences have been associated, both naturally and experimentally, with certain tumours. In the pro-viral LTR from multicentric lymphomas a certain 21 base pair tandem repeated sequence has been found which not only modulates LTR driven gene expression but also confers a replicative advantage. Duplicated enhancers in pro-viral LTR have been discovered to be uniquely associated with T-cell derived lymphomas, particularly thymic lymphomas. Route of inoculation and the host itself are other factors that affect the course of disease.

Research within the subject is ongoing and hopefully the information gained will not only help FeLV-infected cats but can be used when treating tumours of a different cause, or an animal of a different breed. Further knowledge of FeLV might also help gain insight into the mechanisms of other retroviruses, such as the Human Immunodeficiency Virus (HIV).
