Abstract

Anthelmintikagruppen bensimidazoler (BZ) har använts på många djurslag sedan början av 1960-talet. BZ verkar genom att hindra αβ-tubulin dimerer att bygga på mikrotubuli. Cellernas cytoskelett bryts då ner vilket hämmar många viktiga funktioner som exempelvis förmågan till celldelningar. Att BZ är lågt toxiska mot värddjuret har gjort dem populära, men den frekventa användningen har lett till resistensutveckling hos många nematodarter. De nematoder där resistensen är mest utbredd är Haemonchus contortus hos får och inom gruppen cyathostomer hos häst. Det finns i dagsläget allt färre besättningar där BZ har tillräcklig effekt mot dessa parasiter. För att motverka fortsatt resistensutveckling behövs mer kunskap om de underliggande mekanismerna. I denna litteraturgenomgång har nuvarande kunskap om resistensmekanismerna mot BZ hos H. contortus och cyathostomerna sammanställts för att ge en överblick av vad vi vet idag. Mekanismerna bakom resistensen hos H. contortus är de mest studerade och där har en mutation i kodon 200 i β-tubulin isotyp 1-genen visats vara korrelerad till resistensen. Mutationen, F200Y, gör att tyrosin uttrycks istället för fenylalanin vilket minskar antalet bindningar mellan tubulin och BZ. Hos maskar som saknat mutationen i kodon 200 har istället en mutation i kodon 167 eller 198 upptäckts och även dessa anses vara kopplade till resistensen hos H. contortus. Hos hästens cyathostomer är inte kopplingen mellan mutationen F200Y och resistensen lika stark som hos H. contortus och det har visats att resistenta isolat kan sakna denna mutation. Samma substitution som setts i kodon 200 har här setts i kodon 167 så att den då kodat för tyrosin istället för fenylalanin och detta verkar vara kopplat till fenbendazolresistens. Även om framsteg har gjorts inom området så krävs mer forskning för att helt förstå mekanismerna bakom resistensen mot bensimidazoler.

The benzimidazole (BZ) anthelmintics have since the early 1960’s been used in a lot of different species for treatment of gastrointestinal parasites. They inhibit the dimer of αβ-tubulin to attach to microtubules which cause depolymerisation of the microtubules and the cytoskeleton is degraded. This affects many essential functions in the cell, resulting in death of the parasite. The low toxicity of BZ to the host made them popular but frequent use has led to resistance in many nematode species. The resistance is most widespread among Haemonchus contortus in sheep and cyathostomins in horses. Nowadays, there are no farms where benzimidazole is still acceptably effective against the above mentioned nematodes. Knowledge about the underlying mechanisms of resistance is needed to develop effective anhelmintic drugs. In this literature review the present knowledge about the mechanisms of BZ-resistance in H. contortus and the cyathostomins have been compiled to give an overview of what we know today. The resistance mechanisms are most studied in H. contortus where a mutation in codon 200 in the β-tubulin isotype 1 gene has been shown to be strongly correlated to the resistance. This mutation, F200Y, leads to an expression of tyrosine instead of phenylalanine which is considered to decrease the number of bonds between BZ and β-tubulin. In worms that lack the mutation in codon 200, mutations in codon 167 or 198 have been identified and these are also considered to be correlated to the resistance. The link between the mutation F200Y and resistance is not as strong in the cyathostomins as in H. contortus and it has been shown that some resistant isolates even lack this mutation. The same substitution, which has been seen in codon 200, has instead been detected in codon 167 and this seems to be correlated to fenbendazole resistance in the cyathostomins. Although great insights have been gained more research is needed to fully understand the underlying mechanisms of resistance.
