Abstract

Feromoner är substanser som används vid kemisk kommunikation mellan individer av samma art. Hos häst har ännu inga feromoner identifierats men studier gjorda i Litauen tyder på att kresoler, ämnen som bildas i tarmen vid nedbrytning av bland annat aminosyror och som utsöndras via urin och avföring, kan ha feromon effekt hos häst.
Syftet med detta examensarbete var att undersöka förekomsten av kresoler och andra substanser i urin och brunstsekret från ston i östrus med hänsyn till variation mellan dagar i brunst och mellan provmaterial.
Prover samlades in från 8 ston under 10 brunster och vid tre tillfällen från ston i diöstrus. Uppsamling av flyktiga substanser från urin- och sekretproverna gjordes med ”Solid Phase Micro Extraction” (SPME). För separering och identifiering av substanserna användes gaskromatografi och masspektrometri. De relativa mängderna p- och m-kresol i urin och sekret beräknades utifrån kromatogram för varje dag i brunst och jämfördes mellan dagar i brunst och mellan urin och sekret.
P- och m-kresol förekom i både urin och sekret. Generellt innehöll sekret större mängder kresol än urin. I sekret var nivåerna av både p- och m-kresol lägre dag 3 före ovulation än övriga dagar med en stegring mot ovulationsdagen. Mängderna kresol i urin varierade under hela brunsten och ingen tydlig stegring närmare ovulation kunde ses. Diöstrusurin innehöll något mindre p-kresol än östrusurin, och i stort sett ingen m-kresol.
Både urin och sekret visade sig innehålla ett stort antal ämnen utöver kresoler. Det gick inte att utifrån en översiktlig genomgång av kromatogrammen avgöra om det förelåg någon skillnad mellan provtyper och mellan dagar i brunst.

Resultaten tyder på att både urin och brunstsekret från ston innehåller information om reproduktionsstadium och att p - och m-kresol kan vara de ämnen som förmedlar denna information, var för sig eller tillsammans med andra ämnen som finns i urin och sekret.

Pheromones are substances that are used in chemical communication between individuals of the same species. So far, no pheromones have been identified in the horse. Studies done in Lithuania suggested that cresols, substances that are produced in the intestines during for example amino acids degradation and excreted in urine and faeces, could have pheromonal effect in horses.
The purpose of this study was to investigate the presence of cresols and other substances in urine and oestrous secretions from mares in oestrus and to investigate whether there were any variations between days of oestrus and between type of sample.
Samples were collected from 8 mares during 10 oestruses and in three cases from dioestrous mares. Collection of volatile substances from urine and oestrous secretions were done with ”Solid Phase Micro Extraction” (SPME). Gas chromatography and masspectronomy were used for separatation and identification of the substances. The relative amounts of p- and m-cresol in urine and secretions were calculated from chromatograms for each day of oestrus and were then compared between days of oestrus and between urine and secretions.
P- and m-cresol were present in both urine and secretions. Generally, oestrous secretions contained a higher amount of cresol than did urine. The levels of both p- and m-cresol in secretions were lower on day 3 before ovulation than other days of oestrus with an increase towards the day of ovulation. The amounts of cresols present in urine varied during the whole oestrous period and no obvious increase towards ovulation could be seen. Dioestrous urine contained slightly less p-cresol than did oestrous urine, and almost no m-cresol.
Both urine and secretions contained a large number of substances apart from cresols. Without calculating the peaks of these other substances in the chromatograms it was not possible to determine whether there were any differences between sample types and days of oestrus.

The results suggest that both urine and oestrous secretions contain information about reproductive status and that p- and m-cresol may be the substances that convey this information, by themselves or together with other substances found in urine and oestrous secretions.
