Abstract

Timolol är en icke-selektiv betareceptorblockerare som huvudsakligen används för att sänka det
intraokulära trycket hos människor och djur. Trots topikal användning absorberas timolol i systemcirkulationen och ger effekter kopplade till blockering av det sympatiska systemet. Sänkt hjärtfrekvens är den mest uttalade systemiska effekten hos människa. Huruvida oftalmisk timolol ger
systemiska bieffekter på häst är ej känt.
I försöket behandlades 8 hästar med 0,2 ml oftalmisk timolol 0,5 % morgon och kväll fem dagar i
rad och hästarnas hjärtfrekvens i vila monitorerades både före och efter läkemedelsadministreringen. Dessutom utfördes en motsvarande men kortare mätning där placebo administrerades i
stället för timolol. Det erhållna resultatet visar en signifikant minskning av hästens hjärtfrekvens
50 och 60 minuter efter administrering av timolol ögondroppar. Detta indikerar en sannolik
systemisk respons vilket bör iakttas vid både medicinska indikationer samt vid
tävlingssammanhang.

Timolol is a nonselective β-blocker that is predominantly used to treat high intraocular pressure in
humans and animals. Despite topical use, timolol enters the systemic circulation and affects the
body by diminishing the sympathetic activity. Decreased heart rate is the most pronounced
systemic effect in humans. Whether ophthalmic timolol causes systemic side effects in the horse is
not known.
Eight horses were administered 0.2 ml ophthalmic timolol 0.5% solution morning and evening for
five consecutive days and the heart rate in rest was monitored both before and after drug
administration. In addition, a corresponding but shorter measurement was performed when
placebo was administered instead of timolol. The obtained result indicates a reduction in heart rate
50 and 60 minutes after the administration of timolol. This suggests a likely systemic response
which should be considered when using timolol in glaucomatous horses. It also suggests that the
performance of the horse is likely to be affected.
