Abstract

Hos hundrasen berner sennen har rasklubben de senaste åren observerat en ökning av fruktsamhetsrelaterade problem, såsom att tikar inte blir dräktiga, minskat antal valpar i kullarna och förlossningsproblem. Dräktighetsprocenten hos berner sennen ligger idag 10–20 % lägre än hos andra raser.
Denna problematik kan på sikt leda till en för snäv avelsbas, som är potentiellt skadligt för rasen
med hänsyn till ökad risk att utveckla andra genetiska sjukdomar och defekter som förekommer i en
hundras på grund av minskad genetisk variation.
Syftet med denna studie var därför att undersöka möjliga ärftliga faktorer till fruktsamhetsproblematik hos hanhundar med målsättningen att finna en gemensam nämnare i stamtavlorna hos
de undersökta hundarna och potentiellt finna en mutation i någon del av genomet som går att koppla
till den nedsatta fruktsamheten.
Blod- och spermaprover samlades in från 65 friska berner sennenhanar och ett blodprov från en tik.
Spermaproverna analyserades sedan med avseende på totalantal spermier, motilitet och morfologi.
Normalvariationen av spermiesvansdefekter hos friska hundar är <20 %. Grundat på detta
identifierades en avvikande fenotyp i form av ökad andel (>20 %) svansdefekter än den som
vanligtvis förekommer hos en frisk hund. Baserat på denna fenotyp undersöktes samtliga hundars
härkomst i minst åtta generationer för att försöka lokalisera en gemensam nämnare hos individerna
med fenotypen. Detta gjordes ur flera hänseenden; utifrån olika gemensamma anfäder, geografiska
födelseplatser och inavelsgrad. Resultatet visade en ökad andel svansdefekter hos individer med fler
svenskregistrerade mor- & farföräldrar jämfört med hundar med inga eller få svenskregistrerade
mor- & farföräldrar. Detta tyder på både en rasbunden och en populationsbunden problematik vilket
talar för att defekten har ärftliga riskfaktorer.
PCR-analys följt av Sanger-sekvensering utfördes på åtta av individerna för att undersöka
allelfrekvensen vid positionen av två sedan tidigare kända mutationer; en mutation i genen DNAH1,
som har setts hos människa kopplat till den icke-syndromiska spermiedefekten Multiple Morphological Abnormalities of the sperm Flagella (MMAF), som leder till infertilitet hos män, samt en
mutation i genen SFXN5 som ospecifikt har länkats till spermiemorfologi i den turkiska hundrasen
aksaray malaklisi. För DNAH1 genen var resultaten inte bedömningsbara på grund av fel i PCRprocessen. För SFXN5 genen påvisades ingen genetisk variation mellan individerna vilket talar för
att genen inte är länkad till den studerade fenotypen.
Ytterligare forskning i ämnet behövs för utökad kunskap om fertilitetsproblem i berner sennenpopulationen i världen.

The Swedish breed association for Bernese mountain dogs has observed an increase in fertilityrelated problems such as low pregnancy rates, reduced litter sizes and an increased number of
dystocia during the recent years. The pregnancy rate of the Bernese mountain dog is currently 10–
20% lower than in other breeds. This problem can eventually lead to a reduction of potential dames
and sires used for breeding and a narrowed genetic basis, harmful to the breed when considering
increased risk to develop other potential genetic diseases and defects.
The purpose of this study was therefore to investigate possible hereditary factors for fertility
problems from the male perspective with the aim of finding a common denominator in the pedigrees
of the examined dogs and potentially finding a mutation in some part of the genome that can be
linked to the reduced fertility.
Blood and semen samples were collected from 65 healthy Bernese mountain dog males and blood
from one female, all registered in Sweden. The semen samples were analysed for total number of
sperm, motility and morphology. Healthy dogs normally have <20% sperm tail defects. Based on
this, a phenotype was identified in the form of an increased proportion (>20%) of tail defects
compared to what usually occurs in a healthy dog. Based on this phenotype, the descent of all dogs
was examined in pedigrees for at least eight generations in an attempt to locate a common hereditary
denominator in the individuals with the phenotype. This was done using several parameters; based
on different common ancestors, geographical places of birth and level of inbreeding.
The results showed an increased proportion of tail defects in the individuals with more Swedishregistered grandparents compared with dogs with no or few Swedish-registered grandparents. This
indicates both a breed-related and a population-related problem, which suggests that the defect is
hereditary.
PCR analysis followed by Sanger Sequencing was performed in eight of the individuals to examine
the allele frequency at the position of two previously known mutations leading to sperm tail defects;
one mutation in the DNAH1 gene, which has been seen in humans linked to the non-syndromic
sperm defect Multiple Morphological Abnormalities of the sperm Flagella (MMAF) leading to
infertility in men, and one mutation in the SFXN5 gene which has been linked to sperm morphology
in the Turkish dog breed Aksaray malaklisi. For the DNAH1 gene, the results were inconclusive due
to errors in the PCR process. For the SFXN5 gene, no genetic variation was detected between the
individuals, which indicates that the gene is not linked to the studied phenotype.
Further research is needed to investigate the fertility problems in the Bernese mountain dog
worldwide.
