Abstract

Invasiva främmande arter kan ge negativa effekter på den biologiska mångfalden och våra naturliga
ekosystem. Parkslide, Reynoutria japonica är en invasiv främmande art som anses vara mycket
svårbekämpad. Den är väldigt spridningsbenägen med sina kraftiga jordstammar (rhizom) som
bildar stora sammanhängande bestånd. Syftet med mitt arbete var 1) att studera om rhizom av
parkslide hade dött eller fortfarande levde efter värmebehandling, och 2) att undersöka om
infärgning av propidiumjodid (PI) kunde hjälpa till att avgöra om materialet fortfarande var vid liv.
Värmebehandling på 80˚C som pågick åtta dygn visade sig vara en effektiv metod att döda rhizomen
då PI parallellt med infärgning med fluoresceindiacetat (FDA) inte visade på några livstecken i de
behandlade rhizomen. Däremot rhizomen som hade värmebehandlats 80˚C i ett dygn visade med PI
få cellkärnor och med FDA syntes en tydlig fluorescens från cytoplasman, vilket indikerar att
rhizomen fortfarande är vid liv. Infärgning med propidiumjodid visade sig vara ett värdefullt
komplement till infärgning med fluoresceindiacetat för att bekräfta vitaliteten hos rhizom. Arbetet
visar att det krävs mer än ett dygn av värmebehandling men det skulle vara värdefullt i framtida
studier att undersöka om tiden kan kortas ner från åtta dygn och ändå vara effektiv.

Invasive alien species can have a negative impact on biodiversity and our natural ecosystems.
Japanese knotweed, Reynoutria japonica is an invasive alien species that is very difficult to
eradicate. It’s very prone to spread with its powerful rhizomes that can form extensive coherent plant
formations. The purpose of this study was to 1) examine if rhizomes from Japanese knotweed has
died or are still alive after heat treatment, and 2) examine if using propidium iodide (PI) can help in
examining if the rhizomes are still alive. Treatment with 80˚C that went on for eight days was
effective in killing the rhizomes based on that PI and FDA revealed no signs of life. On the other
hand, treatment with 80˚C for one day PI showed just a few visible nucleus and FDA showed a lot
of fluorescence from staining the cytoplasm which indicates that the cells live. Using propidium
iodide proved to be helpful in the evaluation of the rhizomes vitality together with fluorescein
diacetate. This study suggests that it takes longer than one day of heat treatment to kill the rhizomes
from Japanese knotweed but would be worthwhile in future studies to see if eight days can be
shortened and still be effective.
