Abstract

Anti-Müllerskt hormon (AMH) har föreslagits som en biomarkör för spermakvalitet, eftersom hormonet produceras av sertolicellen som stödjer spermiernas utveckling. Förändringar i produktion av
AMH antas kunna visa på nedsatt funktion i sertolicellen, och på så sätt nedsatt funktion i spermieutvecklingen. Hittills har ingen forskning om AMH som kvalitetsmarkör för sperma utförts på hundar. Som en del i ett större projekt undersöks i det här arbetet om koncentrationer av AMH i serum
och seminalplasma kan kopplas till andra markörer för spermakvalitet. I den här studien utfördes en
andrologisk undersökning inkluderande ett spermaprov och ett blodprov för hormonanalys på 65
hanhundar av rasen berner sennen. Multipel linjär regressionsanalys utfördes för att undersöka samband mellan koncentrationen av AMH och olika parametrar hos hundarna. Vi fann att koncentrationen av AMH i serum stiger med hundarnas ålder. Dessutom var AMH-koncentrationen kopplad
till spermiemotilitet och andel spermier med defekt morfologi, och specifikt spermier med huvuddefekter. Högre koncentrationer av hormonet i serum var associerat med lägre motilitet och fler
morfologiskt defekta spermier. Vi fann ingen koppling mellan koncentrationen av AMH i serum
och testikelstorleken eller det totala antalet spermier per ejakulat. Hos 16 av hundarna undersöktes
koncentrationen av AMH i seminalplasma, och hormonet var odetekterbart i alla 16 prover. Sammantaget finns indikationer på att koncentrationen av AMH i serum kan användas som markör för
spermiemotilitet och andelen spermier med defekta huvudformer.

Anti-Müllerian hormone (AMH) has been proposed as a biomarker for semen quality, since the
hormone is produced by the Sertoli cell which also supports the development of sperm. Changes in
production of AMH are suggested to predict decreased function in the Sertoli cell, and by that a
decreased function in sperm development. To this date, there is no published research investigating
if AMH could be used as a marker of quality of sperm in dogs. In the present study, as a part of a
project investigating fertility in male Bernese mountain dogs, concentrations of AMH were measured in serum and seminal plasma and correlated to parameters of quality of sperm. In this study
65 male Bernese mountain dogs were andrologically examined, including semen sampling and a
blood sample for hormone analysis. Multiple linear regression analysis was performed to analyse
correlations between concentrations of AMH and various parameters in the dogs. We found that the
AMH concentration increases with the age of the dogs. Concentration of AMH in serum was correlated to sperm motility and percentage of sperm with defect morphology, specifically defects in the
head region of the sperm. High concentrations of AMH in serum was associated with lower percentage of motile sperm and higher percentage of defect sperm. We were not able to find any correlation
between serum concentrations of AMH and testicular volume or total number of sperm per ejaculate.
In 16 of the dogs concentrations of AMH were measured in seminal plasma, and the hormone was
undetectable in all 16 samples. In conclusion there are indications that AMH could be used as a
biomarker for sperm motility and percentage of sperms with defects in the head region.
