Abstract

Caprin artrit encefalit (CAE) och låga koncentrationer av αS1-kasein (αS1-kn) i mjölken är två problemområden inom den svenska getproduktionen. Caprin artrit encefalit är en virussjukdom som liknar humant immunbristvirus (HIV) hos människa och maedi visna hos får. Sjukdomen går inte att behandla och utslagning av hela besättningen är en vanlig metod för att förebygga vidare smitta. En mutation i genen CCR5 som ger resistens mot sjukdomen CAE har hittats hos getter och genotyperna T'T'/C'T' har visats vara kopplade till en högre förekomst av virus jämfört med genotypen C'C'. Det är intressant att se om denna mutation förekommer i den svenska getpopulationen och syftet med studien var att hitta genetisk variation i CCR5-genen. Ett ytterligare syfte var att jämföra variationen med kliniska data för CAE för att se om samband mellan genotyp och CAE-förekomst finns.
Det andra problemområdet berör ostutbytet från mjölk vilket påverkas av αS1-kn-koncentrationen. Låga koncentrationer resulterar i ett lägre ostutbyte, kg ost per kg producerad mjölk. En mutation i den αS1-kn-kodande genen CSN1S1 kallad den norska deletionen, har visats ha en hög prevalens hos svenska getraser och påverkar syntesen av αS1-kn. Mutationen existerar i tre polymorfa former, A', G' och D', och homozygoter för deletionen, D', har ingen syntes av αS1-kn. Syftet för detta problemområde var att undersöka förekomsten av mutationen hos den Svenska lantrasgeten samt att se om det fanns andra variationer i genen som påverkar αS1-kn-syntesen och därmed koncentrationen av αS1-kn.
Studien analyserade sekvenser från generna CCR5 och CSN1S1 för att hitta genetisk variation hos svenska getter. För CCR5 analyserades sekvenser från 85 Svenska lantrasgetter och 11 Lappgetter för att hitta genetisk variation. Sambandet mellan CAE-förekomst och genotyp undersöktes utifrån data från tre av besättningarna och inkluderade 48 Svenska lantrasgetter. För att studera genetisk variation i CSN1S1 analyserade sekvenser från 48 Svenska lantrasgetter.
Resultaten visade att det fanns två mutationer i CCR5 hos båda getraserna. Mutationerna bestod av tre genotyper vardera T'T', C'C' och C'T', vilken är mutationen som kan kopplas till ökad resistens, och A'A', C'C' och C'A'. C'C'-genotypen visades ha låg förekomst i båda populationerna och det gick inte att hitta något samband mellan genotyp och CAE-förekomst. Förekomsten av deletionen i CSN1S1 var hög och 77% av populationen bar på deletionen. Analysen av sekvensen för CSN1S1 visade ytterligare 12 platser för mutationer och dessa kunde kopplas till G’-allelen för den norska deletionen. Mest intressanta var en extra sekvens på fyra baser "AAAG" som avläses före den norska deletionen i sekvensen, vilken bidrar till en förskjutning i läsramen för proteinet.
Slutsatsen är att förekomsten av ogynnsamma mutationer i CCR5 och CSN1S1 är hög hos raserna Svensk lantrasget och Lappget. Den genetiska variationen visades vara låg och en tydlig LD kunde ses för mutationerna i CCR5 när hela populationen för Svensk lantrasget och Lappget studerades. Studien kunde inte visa på några samband mellan genotyp och förekomst av CAE vilket var förväntat då endast en individ med C’C’-genotypen var tillgänglig för beräkningen. För att beräkningen för genetiskt samband ska bli mer tillförlitligt behövs därför fler genotyper med C’C’ inkluderas.

Caprine arthritis encephalitis (CAE) and low concentrations of αS1-casein (αS1-cn) in the milk are two areas of problem within the Swedish goat production. Caprine arthritis encephalitis is an immunodeficiency virus disease like the human immunodeficiency virus disease HIV and the maedi visna virus in sheep. The disease cannot be treated and when a disease outbreak occurs the most effective preventive measure is to slaughter the whole herd. A mutation in the CCR5 gene that give resilience to CAE have been found in goats and the genotypes T’T’/C’T’ was connected to a higher proviral load compared to the C’C’ genotype. It is interesting to see if this mutation also occurs in the Swedish goat population and the aim was to find genetic variation in the CCR5 gene. Another aim was to compare these variations with clinical data on CAE prevalence to find relationship.
The other area of problem concerns the cheese making properties of the milk which are influenced by the concentrations of αS1-cn. Low concentration results in a lower cheese yield, kg cheese per kg milk produced. A mutation in the αS1-cn coding gene CSN1S1 called the Norwegian deletion, have been found at high prevalence in the Swedish goat breeds and affect the synthesis of αS1-cn. The mutation exists in three polymorphic forms, A’, G’ and D’, and homozygotes for the deletion, D’, have no synthesis of αS1-cn. The aim for this area of problem was to investigate the prevalence of the mutation in the Swedish landrace goat breed and to see if there were other genetic variations found in the gene that could alter the synthesis of αS1-cn and thereby the αS1-cn concentration.
The study evaluated sequences of the two genes CCR5 and CSN1S1 to find genetic variation in the population of Swedish landrace goat. For CCR5 sequences from 85 Swedish landrace goat and 11 from the Swedish breed Lappget was evaluated for genetic variation and 48 Swedish landrace goats was evaluated for CSN1S1. From three of the herds 48 individuals of Swedish landrace goat was used for the evaluation of relationship between CAE prevalence and specific genotype.
The results showed two mutations in the CCR5 gene in both goat breeds. Each mutation consisted in three different genotypes T’T’, C’C’ and C’T’, which was the same mutation connected to resistance, and A’A’. C’C’ and C’A’. The C’C’ genotype was found at very low frequencies in both populations and the result from the relationship study did not find any relationship between genotype and CAE prevalence. The prevalence of the deletion in the CSN1S1 gene var found to be high and 77% of the population carried one allel for the deletion. The evaluation of the sequence showed 12 additional sites for mutations that could be connected to de G’ allele of the Norwegian deletion. The most interesting was an insertion of four bases “AAAG” occurring before the reading site of the Norwegian deletion which will cause a shift in the reading frame of the protein.
The conclusion is that the prevalence of the unfavorable mutations in CCR5 and CSN1S1 is high in the Swedish landrace goat and in Lappget. The genetic variation was shown to be low and a clear LD was seen for the mutations in CCR5 when the entire population of Swedish landrace goat and Lappget was studied. The study did not show any relationship between genotype and the prevalence of CAE, this was to be expected as only one individual with the C’C’ genotype was available for the calculation. For the result about relationship to be more reliable, more individuals with the C’C’ genotype should be included.
