Abstract

Sindbisvirus (SINV) är ett positivt enkelsträngat RNA virus tillhörande genus Alfavirus inom familjen Togaviridae. SINV finns i många delar av världen och orsakar sjukdom hos människor i bland annat Sverige, Finland och Ryssland. I Sydafrika har viruset även kopplats samman med sjukdom hos hästar. SINV cirkulerar i en enzootisk cykel mellan fåglar och myggor och i Sverige finns viruset endemiskt i myggpopulationen i de centrala delarna av Sverige. Eventuell förekomst och sjukdomsbild hos hästar i Sverige är dock okänd. Huvudsyftet med studien var därför att undersöka förekomst av antikroppar mot SINV hos hästar i Sverige. I studien undersöktes även förekomst av antikroppar hos häst mot tick-borne encephalitis virus (TBEV), West Nile virus (WNV) och Usutuvirus (USUV) vilka är virus tillhörande genus Flavivirus inom familjen Flaviviridae. TBEV finns i Sverige men seroprevalens och eventuell sjukdomsbild hos hästar är okänd. WNV och USUV har ännu inte rapporterats hos hästar i Sverige men antikroppar mot dessa virus har hittats hos hästar i flera länder i Europa.

Provmaterialet som består av hästserum är insamlat från klinisk kemiska laboratoriet på universitetsdjursjukhuset (UDS) vid Sveriges lantbruksuniversitet under perioden april-augusti. Proverna kommer antingen från patienter på hästkliniken på UDS eller är inskickade från veterinärer runtom i landet. Totalt samlades 651 prover in och av dessa analyserades 633 för SINV E1, 187 för SINV E2 samt 614 för TBEV, WNV och USUV. Det som undersöktes var förekomst av IgG antikroppar. Proverna analyserades i en suspension multiplex immunoassay (SMIA) och resultatet från denna erhölls sedan genom analys i ett Luminex-Magpix instrument.

Utifrån de cut-off värden som beräknades var totalt 78 prover SMIA positiva för SINV vilket motsvarar 12 % av antalet analyserade prover, 216 prover var SMIA positiva för TBEV vilket motsvarar 35 % av antalet analyserade prover. För WNV och USUV var 73 respektive 24 prover SMIA positiva vilket motsvarar 12 % respektive 4 % av antalet analyserade prover. De insamlade proverna grupperades efter län och hade en spridning från Norrbottens län i norr till Skåne län i söder. SMIA positiva prover för SINV och TBEV kom från 17 olika län och för WNV och USUV-positiva kom proverna från 11 respektive 10 olika län. Positiva SMIA prover fanns för samtliga virus från alla månader prover samlats in.

Resultatet i denna studie visar att hästar från många delar av Sverige eventuellt utsatts för SINV infektion vilket är av intresse då detta tidigare endast rapporterats från Sydafrika. Resultatet visar även att hästar i Sverige eventuellt utsatts för infektion med TBEV, WNV och USUV då det fanns SMIA positiva prover även för dessa virus. Dessa resultat är dock ganska osäkra, särskilt de för WNV och USUV, och behöver konfirmeras med ett plaque reduction neutralization test (PRNT) för påvisande av neutraliserande antikroppar för respektive virus. Om positiva prover hittas även med PRNT är det även av intresse att samla in information om eventuella symtom som de berörda hästarna har uppvisat för de olika virusen.

Sindbis virus (SINV) is a positive single-stranded RNA virus belonging to the genus Alfavirus in the family Togaviridae. SINV is found in many parts of the world and causes illness in people in Sweden, Finland and Russia, amongst other countries. In South Africa, the virus has also been linked to disease in horses. SINV circulates in an enzootic cycle between birds and mosquitoes and in Sweden the virus is endemic in the mosquito population in the central parts of Sweden. However, possible incidence and clinical picture among horses in Sweden is unknown. The main aim of this study was therefore to investigate the presence of antibodies against SINV in horses in Sweden. The study also investigated the presence of antibodies in horses against tick-borne encephalitis virus (TBEV), West Nile virus (WNV) and Usutu virus (USUV) belonging to the genus Flavivirus in the family Flaviviridae. TBEV is present in Sweden but the seroprevalence and possible clinical picture in horses is unknown. WNV and USUV has not yet been reported in horses in Sweden but antibodies against these viruses has been found in horses in several countries in Europe.

The sample material, consisting of horse serum, was collected from the clinical chemical laboratory at the university animal hospital (UDS) at Swedish University of Agricultural Sciences during the period April-August. The samples came either from patients at the horse clinic at UDS or were sent from veterinarians around the country. A total of 651 samples were collected, of which 633 were analysed for SINV E1, 187 for SINV E2 and 614 for TBEV, WNV and USUV. What was investigated was the presence of IgG antibodies against the different viruses. The samples were analysed in a suspension multiplex immunoassay (SMIA) and the results from the SMIA were then obtained by analysis in a Luminex-Magpix instrument.

Based on the cut-off values calculated, a total of 78 samples were SMIA positive for SINV which corresponds to 12% of the samples analysed, 216 samples were SMIA positive for TBEV which corresponds to 35% of samples analysed. For WNV and USUV 73 respectively 24 samples were SMIA positive which corresponds to 12% and 4% of the samples analysed. The collected samples were grouped by county and had a spread from Norrbotten county in the north to Skåne county in the south. SMIA positive samples for SINV and TBEV came from 17 different counties and for WNV and USUV the samples came from 11 and 10 different counties respectively. Positive SMIA samples were present from all the months that samples were collected.

The result of this study shows that horses from many parts of Sweden may have been exposed to SINV infection, which is of interest since this previously only has been reported from South Africa. The result also shows that horses in Sweden may have been infected with TBEV, WNV and USUV as there were positive SMIA samples for these viruses as well. However, these results are quite uncertain, especially those for WNV and USUV, and need to be confirmed by a plaque reduction neutralization test (PRNT) to demonstrate the presence of neutralizing antibodies for each virus respectively. If positive samples also are found with the PRNT, it is also of interest to gather information about possible symptoms that the horses involved have shown
for the different viruses.
