Abstract

Bedömningar av reproduktionstoxicitet görs på både tama och vilda djur, ibland används djurarter som så kallade indikatorarter. En indikator kan beskrivas som en icke-mänsklig organism vilken kan reagera på en miljöförorening innan föroreningen påverkar människor. Konceptet är inte nytt och används i syfte att förvarna människan innan föroreningen kommer att drabba den humana populationen eller bli ett miljömässigt problem. Användandet av indikatorarter ger också en uppfattning över artens reproduktiva förmåga och dessa djurarter kan givetvis även studeras enbart i syfte för artens egen skull. Studier på vilda djur står inför flera praktiska problem och tiden mellan avlivning och provtagning utgör en betydande faktor för kvaliteten på proverna. Detta påverkar inte bara vilka undersökningar som kan genomföras utan också resultaten av dessa.

Även om vävnaden förvaras under så optimala förhållanden som möjligt så kan den till slut komma att påverkas. Temperatur och tid är två faktorer som har visat sig spela betydande roll för förvaring av testikelvävnad. Hur stor deras påverkan på vävnaden är och hur detta i sin tur påverkar olika typer av undersökningar är ännu inte helt fastställt. Aspekterna tid och temperatur har därför varit av intresse i denna studie. Studiens mål var att simulera ett antal suboptimala fältlika förhållanden med olika temperatur och under olika långa förvaringsperioder. En vanligt förekommande metod för utvärdering av hanlig reproduktions-toxicitet vid experimentella studier är kvantifiering av homogeniseringsresistenta spermatider. Denna metod har valts i denna studie och 68 vävnadsprover ifrån testiklarna till en tjur har homogeniserats vartefter antalet spermatider har räknats manuellt med hjälp av en räknekammare.

Resultaten tydde på att kylförvaring av testikeln under några dagar innan kvantifiering är möjlig. Även förvaring i rumstemperatur är möjlig inom ett par dygn. Proverna kan också frysas om direkt bedömning inte är genomförbar, infrysningen kan ske direkt eller efter kylförvaring men frysta prover som har tinats bör inte frysas på nytt för att användas vid ett senare tillfälle. Frysta prover hade inte signifikant lägre antal homogeniseringsresistenta spermatider, men medelvärdena låg konstant lägre jämfört kylförvarade prover, vilket är ett observandum om metoden ska användas på arter med känsligare spermatider än tjur. Då spermatiderna är fortsatt motståndskraftiga mot homogenisering trots förvaring i olika temperaturer över tid så har metoden goda förutsättningar att fungera på vilda djur.

Reproduction toxicity assessments are made on both domestic and wild animals, sometimes animal species are used as so-called indicator species. An indicator can be described as a nonhuman organism which can respond to an environmental pollution before pollution affects people. The concept is not new and have long been used to warn humans against contamination that will affect the human population or become an environmental problem. The use of indicator species also gives an idea of the species own reproductive capacity, and of course these species can also be studied whit this as the only objective. However, studies that include wild animals face several practical problems, the time between death and tissue collection is a significant factor when it comes to the quality of the samples. This does not only affect the type of analysis that can be carried out but also the results of those analyses.

Although the tissue is stored under optimal conditions, eventually it will be affected. Temperature and time are two factors that have been shown to play a significant role in the storage of testicular tissue. The extent of the impact on the tissue and in turn how this affects different types of investigations is not yet fully established. The aspects of time and temperature have therefore been of interest in this study. The aim of this study was to simulate, a number of less optimal, field-like conditions with different temperatures and different storage time. A commonly used method for evaluating male reproduction toxicity in experimental studies is the quantification of homogenization resistant spermatid heads. This was chosen for this study and 68 tissue samples from the testicles of a bull were homogenized, after which the number of spermatids was quantified manually with the help of a counter chamber.

The results indicated that cold storage of the testicle for a few days before quantification is possible. Storage at room temperature is also possible during a couple of days. The samples can also be frozen if direct assessment is not feasible. Freezing can be done directly or after cold storage but frozen specimens that have been thawed should not be frozen again for use at a later date. Frozen specimens did not have significantly lower numbers of homogenization-resistant spermatids, but the mean values were constant lower compared to refrigerated samples, which is an observation if the method is to be applied to species with more sensitive spermatids than bull. As the spermatids are still resistant to homogenization despite storage at different temperatures over time, the method has good possibilities to work on wild animals.
