Abstract

Immunisering mot gonadotropin-releasing hormone (GnRH) har visat sig vara effektivt mot att hämma galtlukt på griskött och oönskat aggressivt beteende bland hangrisar. I ett samhälle där vi sätter allt högre krav på våra produktionsdjurs välfärd är efterfrågan på metoder som
inte innebär smärta eller lidande för djuren stor. Det finns dessutom positiva produktionsrelaterade effekter med immunokastrering gentemot klassisk kirurgisk kastrering,
såsom ökad tillväxtpotential. Faktorer som dessa talar för immunokastrering, men samtidigt finns många svårigheter med att vaccinera mot något kroppseget och således bryta kroppens tolernas mot självt. Den här litteraturstudien behandlar svårigheterna med att rikta ett vaccin mot ett endogent hormon, vad som har lett fram till dagens upptäckter och de eventuella problem som kan uppstå i framtiden. Korsreaktivitet med andra GnRH-isoformer vars
funktion i kroppen inte är helt kända väcker frågor om eventuella bieffekter som ännu inte är upptäckta. Man har framgångsrikt kunnat rikta antikroppssvaret från vaccinationen mot den GnRH-isoform som reglerar reproduktivitet, och man har även identifierat vilka aminosyror i GnRH-peptiden som spelar störst roll i förmågan att väcka ett antikroppsvar mot GnRH.
Tidpunkten för vaccinationen har också betydelse då en tidigare vaccinering ger en ännu effektivare reducering av de könshormoner som man vill hämma. Immunisering mot GnRH
resulterade i 100% av fallen i tillräckligt reducerade könshormoner samt minskat aggressivt beteende. Dock kvarstår fortfarande frågor om risker för biverkningar och långsiktiga effekter som ännu inte har upptäckts.

Immunization against gonadotropin-releasing hormone (GnRH) is effective in reducing boar taint in pork and unwanted aggressive behavior in male pigs. A continuously rising interest in the welfare of our production animals demands for castration methods that doesn’t involve pain or suffering. Moreover, there is positive production related effects in immunocastrated pigs compared to surgically castrated, such as increased growth potential. This puts a positive look on immunocastration, but there is also difficulties involved when the target of the vaccine is something that is supposed to be native and therefore needed to break the tolerance against self. This literature review discusses the difficulties in directing a vaccine against an endogenous hormone, the discoveries that has led up to today’s knowledge and the potential problems that may occur in the future. Cross-reactivity with other GnRH-isoforms whose functions is not fully understood raises questions about possible yet unknown side-effects.
Antibody response has successfully been directed to the preferred isoform of GnRH, and also identified the amino acids in the sequence that are most important for inducing an immune response against GnRH. The timing of the vaccination is also of importance since early
immunization results in more efficient inhibition of the testicular hormones that causes boar taint and sexual behavior in male pigs. Immunization against GnRH was a 100 % effective in suppressing boar taint causing compounds under the threshold values and also to reduce the
sexual behavior. However, there is still questions to be answered about the potential risk for side effects and long-term effects that are not yet discovered.
