Abstract

Studien undersökte om det fanns något samband mellan årstid och resultat på ett antal test för fertilitetsfaktorer.

Att försöka förutse ett handjurs fertilitet är komplext, inte minst då spermieegenskaperna skiljer sig åt mellan hingstar och även mellan ejakulat från samma hingst. Det finns många funktioner som måste analyseras för att säkert bedöma en oprövad hingsts fruktsamhet. Forskningen på området är begränsad och består ofta av studier med litet underlag eller retrospektiva studier där skötselfaktorer och stoegenskaper kan inverka. Som fertilitetsmått används ofta dräktighetsprocent men detta värde kräver stort underlag för att kompensera för eventuella fertilitetsstörningar hos de ston som används och eventuell bristande gynekologisk diagnostik. Mer kontrollerade studier som fortfarande är representativa för populationen är svåra att genomföra då det är svårt att standardisera levnadsförhållanden för sporthästar. Hingstar som används i avel selekteras ofta på prestation varför det är vanligt att de tränas och tävlas parallellt med sin avelskarriär.

I studien analyserades ordinarie transportspermadoser från nio hingstar av halvblodsras. Prover samlades vid fyra tillfällen under två veckor i månadsskiftet april/maj samt vid fyra tillfällen under två veckor i augusti. Två hingstar exkluderades då de lämnade för få prover. Totalt analyserades 42 prover. Proverna analyserades med avseende på koncentration, progressiv motilitet, total motilitet, andel spermier med normal morfologi, viabilitet med SYBR-14 och Hoechst33258, produktion av superoxid (O2-), produktion av väteperoxid (H2O2) samt kromatinintegritet (SCSA). Signifikant effekt av årstid sågs för total motilitet (p = 0,0256); och superoxidproduktion (p ≤0,0246 ). För väteperoxidproduktion sågs en svag effekt (p ≥0,0716). Motiliteten var högre i proverna från augusti jämfört med de tagna i april/maj medan superoxidproduktion var lägre.
Då man ser ökad motilitet i augusti jämfört med april/maj och motilitet har visat god korrelation till fruktsamhet kan man från studien anta att fertiliteten ökar senare på säsongen. Underlaget för studien var begränsat varför försiktighet bör vidtas då resultaten används, särskilt om de skall tillämpas för hingstar av annan ras.

The aim of the study was to determine the effect of season on sperm quality parameters.

The large number of factors influencing the outcome of a mating complicates fertility prediction. The studies available generally have one of two main weaknesses; they are either made retrospectively in which the effects of mare and management factors confound the results or they are made in a more controlled environment but with a limited number of stallions. Sperm quality varies greatly between stallions and even between ejaculates from the same stallion. A large amount of data is thus required to get results that are useful in practice. Most warmblood stallions are bred because of their good sport performance; they are therefore in training and competing during the breeding season which makes it difficult to standardize their management.

In this study chilled fresh semen from nine warmblood stallions was analyzed for known indicators of fertility. The aliquots used in the study were taken from commercial AI (Artificial Insemination) doses. Samples were collected at four times during two consecutive weeks in April and May, which is in the first third of the Swedish breeding season, and then again four times during two weeks in August which is the end of the breeding season. A total of 42 samples were analyzed. Two stallions were excluded because there were only five samples from each. The sperm concentration, total and progressive motility, normal morphology, viability with SYBR-14 and Hoechst 33258, production of superoxide (O2-), production of hydrogen peroxide (H2O2) and chromatin integrity (SCSA) were evaluated. A significant effect of season was seen for total motility (p p = 0,0256) and production of superoxide (p ≤ 0,0246). For hydrogen peroxide production a low significance (p ≥0,0716) was seen. Motility increased in the August samples compared to the samples from April and May, the superoxide production decreased. Other parameters were not significantly affected by season.

The study shows that there is a significant increase in sperm motility by the end of the breeding season. Since motility has been closely linked to fertility one could conclude that stallion fertility may be higher by the end of the breeding season. However, only a restricted number of animals were studied and the result may not be valid for a larger stallion population, particularly for other breeds
