Abstract

Potatisbladmögel är en allvarlig växtsjukdom som orsakas av oomyceten Phytophthora infestans. Denna växtpatogen har både asexuell och sexuell förökning. För att sexuell förökning ska uppstå krävs att de båda parningstyperna, A1 och A2, är närvarande. Sexuell förökning ger upphov till tjockväggiga och persistenta oosporer. Oosporbildningen har i tidigare studier visats bli påverkad av fungicider på olika sätt, men det finns ingen vetenskaplig konsensus för huruvida fungicider ökar eller minskar bildningen av oosporer. I detta försök samlades potatisblad av sorten Kuras som var infekterade av P. infestans in från ett fältförsök, där ett led behandlades enligt ett rekommenderat sprutprogram (K2) och ett led var obehandlat (K0). De insamlade proverna isolerades, parningstypsbestämdes och korsades enligt en korsningsmatris. Alla korsningar gjordes både på agar med inblandad fungicid (Ranman Top, aktiv substans cyazofamid) och på agar utan fungicid.
Resultatet visade att det inte fanns någon skillnad i oosporbildning mellan korsningar gjorda på agarplattor med respektive utan fungicid. Däremot gav korsningar som innehöll minst ett isolat från K2 fler oosporer än korsningar med två isolat från K0, men bara vid odling på agar utan fungicid. Detta indikerar alltså att isolat av P. infestans som överlevt fungicidbehandling i fält bildar mer oosporer när de odlas på artificiellt medium utan inblandning av fungicid. Att K2 inte bildade fler oosporer på agarplattorna med fungicid kan antas bero på en trade-off-effekt mellan högre myceltillväxt och oosporbildning. För framtida studier hade det varit av intresse att även undersöka kopplingen mellan aggressivitet och oosporbildning hos olika isolat av P. infestans.

Potato late blight is a devastating plant disease caused by the oomycete Phytophtho- ra infestans. This pathogen can reproduce both asexually and sexually. For sexual reproduction to take place, both mating types (A1 and A2) must be present. Sexual reproduction results in thick-walled and persistent oospores. In earlier studies, the formation of oospores has been shown to be affected by fungicides in different ways but there is no scientific consensus on how it is affected. In this experiment, potato leaves of the variety Kuras infected by P. infestans were collected from field trial plots treated with fungicides according to a spraying program (K2) and from untreated plots (K0). The samples were isolated, the mating type of the isolates were determined and then crossed according to a matrix. All crossings were done on both agar containing fungicide (Ranman Top, active ingredient cyazofamide) and agar without the fungicide.
The results showed no difference in oospore formation between crossings done on agar with fungicide compared to crossings on agar without fungicide. However, crossings containing at least one isolate from K2 resulted in more oospores than crossings containing two isolates from K0, but only when grown on fungicide free agar. This indicates that isolates of P. infestans that have survived treatment with fungicides in the field, produce more oospores when they are grown on artificial medium free of fungicides. The fact that K2 did not form more oospores on agar treated with fungicide was assumed to be due to a trade-off effect between a higher growth rate of the mycelia and formation of oospores. Future studies could investi- gate the connection between the aggressiveness and ability to form oospores in different isolates of P. infestans.
